Generally, they're not a factor. Few if any of the jobs I applied for asked for transcripts.

I'm still in grad school myself, but I have been on a hiring committee and I was surprised to see that there was no discussion of candidates' grades, even for one candidate who I thought had low-ish grades. We did, however, request transcripts from all candidates.
